Fluorescence microscope uses ultraviolet light as the light source to irradiate the object to be inspected to make it emit fluorescence, and then observe the shape and location of the object under the microscope.
Fluorescence microscopy is used to study the absorption, transport, distribution and localization of substances in cells. Some substances in cells, such as chlorophyll, can fluoresce after being irradiated by ultraviolet rays; some other substances themselves cannot fluoresce, but if stained with fluorescent dyes or fluorescent antibodies, they can also fluoresce after being irradiated by ultraviolet rays. Fluorescence microscope is one of the tools for qualitative and quantitative research on such substances.
The role of the filter
Most fluorescence imaging is done by fluorescence microscopy, which includes the following basic elements:
• Light source: usually xenon arc lamp or mercury lamp, but in recent years also use high-power LED, as well as the use of laser light source
• Filter (incident light): Shrinks the wavelength of incident light to only the wavelength that is left to excite the sample, which is called an excitation filter
• Ddichroic mirror or reflector: reflects the excitation light to the sample and at the same time passes only the emitted light from the sample to the detector
• Filter (emission light): Only pass the emission light wavelength from the sample and block all light passing through the excitation filter, which is called an emission filter as you think
• CCD camera: The emitted light is of no use if it cannot be detected; for fluorescence imaging, the detector is usually a CCD camera, which is also usually connected to a computer screen and can present you with an image.
Fluorescence filters generally comprise a combination of three, an excitation filter, an emission filter, and a dichroic mirror.
